题题目:目: 刺楸种子内含抑制物的活性测定 二二 O O 一二年五月十八日一二年五月十八日 摘摘要要 刺楸(Kalopanax septemlobus)是我国珍贵的国家二级保护植物,它属落叶乔木,适应性 强,抗性强,应用广泛。但是刺楸种子的休眠期极长,萌发条件苛刻,这给林业生产带来 了很大的困难,也限制了该树种的发展,至今仍未找出最佳的解除刺楸种子休眠的方法。 研究刺楸种子内含抑制物的活性大小对研究种子休眠原因和解除种子休眠具有重要意义, 本实验用甲醇和乙醇两种浸提液提取刺楸种子内含抑制物,通过白菜种子生物测定法,以 蒸馏水处理的白菜种子作对照,对不同浓度(0.1g/mL,0.05g/mL,0.025g/mL)浸提液处 理的白菜种子的发芽率、根长和苗高这三个指标进行测定。实验结果表明刺楸种子中含有 大量的抑制物质,且活性较高。0.1g/mL 的甲醇浸提液处理的白菜种子一粒未发,三种处 理的苗高与蒸馏水处理的白菜作比照分别降低了 100%、81.73%、37.6%,根生长量分别降 低了 100%、97.12%、79.24%。乙醇浸提液处理的白菜种子与蒸馏水对照相比,发芽率分 别降低了 8.45%、2.85%、2.11%。0.1g/mL 浓度乙醇浸提液处理的苗高降低了 27.30%, 0.05g/mL 和 0.025g/mL 浸提液处理的却分别增长了 13.78%、1.01%。乙醇浸提液处理的白 菜苗苗根的生长量分别降低 73.92%,11.18%和 2.13%。 关键词关键词刺楸;抑制物;休眠;白菜种子;生物测定 The containing inhibitor activity measure ofThe containing inhibitor activity measure of KalopanaxKalopanax seedsseeds AbstractAbstract:Kalopanax(Kalopanax septemlobus) is our precious plants of the second class state protection, it is a deciduous tree of good adaptability, strong resistance and wide range of applications.But Kalopanax seeds have a very long period of dormancy and seeds germinate under harsh conditions.These have brought great difficulties to forestry production and also limit the development of the species. The of the best lifting Kalopanax seeds dormancy has yet to find out. We use cabbage seed biological determination , the Kalopanax seeds contents were extracted with both methanol and ethanol. We set 0.1g/mL 0.05g/mL, 0.025g/mL three concentration gradient with distilled water as contrast. Under different concentrations of extracts, three indicators of cabbage seeds germination rate, root length and seedling height were measured, and experimental results show that the the Kalopanax seeds contains large amounts of inhibitory substances with high activity. 0.1g/mL methanol extract of cabbage seeds have yet to germinate. Seedling height with distilled water cabbage ck, was respectively reduced by 100%, 81.73%, 37.6%, root growth was reduced by 100%, 97.12%, 79.24%. The ethanol extract of cabbage seeds with distilled water ck, germination rates were reduced by 8.45%, 2.85%, 2.11%. 0.1g/mL concentration of the ethanol extract treatment of seedling height was reduced by 27.30% to the distilled water ck, the 0.05g/mL and 0.025g/mL extract concentration, respectively, have an increase of 13.78%, 1.01%.
