精品文档---下载后可任意编辑 ABCG2在肾癌中的表达及其与肾癌干细胞关系的初步讨论的开题报告 【摘要】ABC运输体谷胱甘肽转移酶2(ABCG2)在多种肿瘤中均有较高的表达,其与肿瘤多药耐药及肿瘤干细胞的关系备受关注。然而,目前关于ABCG2在肾癌中的表达及其与肾癌干细胞关系的讨论还很有限。本讨论将收集肾癌患者组织标本,利用免疫组化及Western blot技术检测ABCG2的表达情况,并利用流式细胞术分离肾癌中的干细胞。通过比较不同临床分期、组织类型及预后情况的肾癌患者ABCG2的表达情况,初步探讨ABCG2在肾癌中的表达特点。并通过原代肿瘤球形体的形成实验及细胞增殖实验证明ABCG2的参加肾癌干细胞特性的调节。估计本讨论能为进一步讨论肾癌发生进展的分子机制提供新思路,并在探究肾癌干细胞特性及相关治疗提供指导意义。 【关键词】ABC运输体谷胱甘肽转移酶2;肾癌;多药耐药;干细胞 【Abstract】 ABC transporter glutathione S-transferase 2 ABCG2 is highly expressed in various tumors, and its relationship with multidrug resistance and tumor stem cells has attracted much attention. However, the current research on ABCG2 expression and its relationship with renal cancer stem cells is still very limited. In this study, renal cancer tissue specimens will be collected, and the expression of ABCG2 will be detected by immunohistochemistry and Western blot techniques. Renal cancer stem cells will be separated by flow cytometry. By comparing the expression of ABCG2 in renal cancer patients with different clinical staging, tissue types, and prognosis, the expression characteristics of ABCG2 in renal cancer will be explored. Through the ation of primary tumor spheres and cell proliferation experiments, it is proved that ABCG2 is involved in the regulation of renal cancer stem cell characteristics. This study is expected to provide new ideas for further research on the molecular mechanisms of renal cancer development, and provide guidance for exploring the characteristics of renal cancer stem cells and related treatments. 【Keywords】 ABC transporter glutathione S-transferase 2; renal cancer; multidrug resistance; stem cells
