APRIL和XIAP在结肠癌、结肠腺瘤及正常结肠黏膜中 的表达及意义 [摘要]目的探讨增殖诱导配体APRIL和X连锁凋亡抑制蛋白 XIAP在结肠癌、结肠腺瘤及正常结肠黏膜组织中的表达及其临床意义。 方法应用免疫组织化学EnVision法检测结肠癌组织标本60例、结肠腺 瘤30例及正常结肠黏膜组织30例中APRIL和XIAP的表达。结果 结肠 癌、结肠腺瘤及正常结肠黏膜组织中APRIL和XIAP的阳性表达率均呈逐 渐降低趋势,3组间APRIL和XIAP的阳性表达率比较,差异有统计学意义 P0.01o结肠癌组织中APRIL的阳性表达与组织分级、淋巴结转移以 及浸润程度密切相关P〈0.01; XIAP的阳性表达与Dukes分期、淋巴结 转移以及浸润程度密切相关P〈0. 01。结论APRIL和XIAP的表达增高 不仅可能在正常黏膜-腺瘤-癌的病理形成过程中起一定作用,更与结肠癌 浸润、转移等恶性生物学行为有密切关系。 [关键词]结肠癌;结肠腺瘤;免疫组织化学;增殖诱导配体;X连锁 凋亡抑制蛋白 [中图分类号]R735. 35 [文献标识码]A [文章编号]1674-4721 2014 09 a -0012-03 Expression and clinical significance of APRIL and XIAP in colon carcinoma, colon adenoma and normal colon mucosa YUAN De-hanl KANG Ju-ling2 SU Yan-tao3 WEN Zhi-xiongl TANG Hua-zhongl 1. Department of Nursing, Shijie Hospital of Dongguan City in Guangdong Province, Dongguan 523290, China [Abstract] Objective To investigate the expression and clinical significance of a proliferation-inducing ligand APRIL and X1 inked inhibitor of apoptosis protein XIAP in colon carcinoma, colon adenoma and normal colon mucosa. s The expression of APRIL and XIAP was detected by immunohistochemical EnVisionTM plus nonhiotin technique in 60 cases of colon carcinoma and 30 cases of colon adenoma, 30 cases of normal colon mucosa. Results The positive expression rates of APRIL and XIAP in colon carcinoma, colon adenoma and normal colorectal mucosa decreased gradually. There was significant difference among the three groups P0. 01 . The expression of XIAP were closely related to Dukes clinical stage, lymph node metascasis and the degree of infiltration P0.01 . The expression of APRIL were closely related to differentiation degree, lymph node metascasis and the degree of infiltration P0. 01 . Conclusion The high expression of APRIL and XIAP may play important roles in the carcinogenesis of colon carcinoma and it is involved in the metastasis and infiltration. [Key words] Colon carcinoma ; Colon adenoma ; Immunohistochemistry; A proliferation-inducing ligand; X-linked inhibitor of apoptosis protein 目前研究已经发现大多数结肠癌的发生都经过了正常黏膜-腺瘤-癌 的过程,但其发生机制至今尚不清楚。近年来一些研究发现,肿瘤的自主 增殖和凋亡失调是结肠癌的“腺瘤-癌演续学说”中的重要机制[1]。增殖 诱导配体a proliferation-inducing ligand, APRIL属于肿瘤坏死因 子超家族新成员,它在消化系肿瘤中的高表达已经引起学者们的广泛关 注。X 连锁凋亡抑制蛋白X-linked inhibitor of apoptosis protein, XIAP是凋亡抑制蛋白inhibitors of apoptosis, IAPs家族中最重 要的caspase抑制物[2]。在体外实验中发现,APRIL表达具有抗凋亡能力, 推测可能与APRIL上调X相联系的XIAP,使caspase-3活性降低有关[3]。 本研究采用免疫组织化学法检测结肠癌、结肠腺瘤及正常结肠黏膜组织中 APRIL和XIAP的表达,旨在进一步阐明正常黏膜-腺瘤-癌的病理机制及两 者在此过程中所起的作用。 1对象与方法 1. 1研究对象 收集2009年8月16日〜2010年2月15日东莞市石碣医院和暨南大 学附属第一医院手术切除结肠癌标本60例、结肠腺瘤30例及正常结肠黏 膜组织30例,均行10甲醛固定,常规脱水,石蜡包埋,4 nm连续切片, 均经病理学证实。 1.2方法 采用免疫组织化学EnVision法检测结肠癌、结肠腺瘤及正常结肠黏 膜组织中APRIL和XIAP的表达,兔抗人多克隆抗体APRIL和XIAP,EnVision 试剂盒、DAB显色试剂盒均购自北京中杉金桥生物技术有限公司。操作程 序严格按照试剂盒说明书进行。
