IGF-1和IGF-1R在无功能性垂体腺瘤中的表达及意义 王志明,蒲九君,孙彦龙,周辉,覃凯,阮伦亮,钟艾凌,杨刚 400016重庆,重庆医科大学附属第一医院 神经外科 [摘要]目的 探讨胰岛素样生长因子1 IGF-1和胰岛素样生长因子1型受体IGF-1R与无功能性垂体腺 瘤侵袭性和复发性的关系。方法60例无功能性垂体腺瘤分为侵袭性复发性组16例、侵袭性非复发性组24例和 非侵袭性非复发性组20例。采用qRT-PCR检测IGF-1和IGF-1R的mRNA表达水平,采用免疫组织化学法和Western blot检测IGF-1、IGF-1R和其下游信号分子血管内皮生长因子VEGF的蛋白表达水平。结果IGF-1和IGF-1R的 mRNA表达水平及IGF-K IGF-1R和VEGF的蛋白表达水平,侵袭性复发性组高于侵袭性非复发性组P0.05,侵 袭性非复发性组高于非侵袭性非复发性组P0.05。相关性分析结果显示,在侵袭性复发性、侵袭性非复发性无 功能性垂体腺瘤组织中IGF-1和IGF-1R蛋白表达量呈正相关r 0.85, r 0.80, P0.05, IGF-1R和VEGF蛋白表达量 呈正相关r 0.74, r 0.70, F0.05;在非侵袭性非复发性无功能性垂体腺瘤组织中IGF-1和IGF-1R蛋白表达量呈 正相关M 0.73, F0.05, IGF-1R和VEGF蛋白表达量无明显相关性F0.05。结论 IGF-1、IGF-1R和VEGF在 侵袭性和复发性无功能性垂体腺瘤中表达上调,三者协调作用,与无功能性垂体腺瘤的侵袭性和复发性恶性行为的 发生、发展可能相关。 [关键词]IGF-1; IGF-1R;无功能性垂体腺瘤;侵袭性;复发性 [基金项目]国家临床重点专科建设项目财社[2011] 170号;重庆市科委自然科学基金CSTC2013jcyjA10079 [通信作者]杨刚,E-mail yg [优先出版] Expression of insulin-like growth factor 1 and insulin-like growth factor 1 receptor in human non-functioning pituitary adenomas correlation with invasiveness and recurrence Wang Zhi Ming, Pu Jiu Jun, Sun Yan Long, Zhou Hui, Qin Kai, Ruan Lun Liang , Zhon Ai Ling, Yang Gang Department of Neurosurgery, First Affiliated Hospital, Chongqing Medical University, Chongqing, 400016, China [Abstract] Objective To investigate the expression levels of insulin-like growth factor 1 IGF-1 and insulin-like growth factor 1 receptorIGF-1R in human non-functioning pituitary adenomas and their relationships with tumor invasion and recurrence in these cases. s Sixty specimens of non-functioning pituitary adenomas were classified into invasive recurrent groups、invasive non-recurrent groups and non-invasive non-recurrent invasive groups. The protein and mRNA expression levels of IGF-1 IGF-1R and VEGF , the downstream molecule of IGF-1R associated signaling pathway, were detected using immunohistochemistry assay, western blot assay and quantitative real-time polymerase chain reaction qRT-PCR, respectively. Results The mRNA expression levels of IGF-1 and IGF-1R and the protein expression levels of IGF-1 IGF-1R and VEGF were significantly higher in invasive non-recurrent group than in non-invasive non-recurrent group P0.05 , and in invasive recurrent groups than in invasive non-recurrent group P0.05.The correlation analysis revealed that there was a positive correlation between IGF-1 and IGF-1R P0.05, IGF-1R and VEGF P0.05 ,in invasive recurrent groups and in invasive non-recurrent group, in non-invasive non-recurrent group, IGF-1 were positively correlated with IGF-lRP0.05 and IGF-1R were not correlated with VEGFP0.05. Conclusion The increased expressions of IGF-1、IGF-1R and VEGF are associated with the invasiveness and recurrence of non-functioning pituitary adenomas.
